The IL-17RA[Biotin]:IL-17A Inhibitor Screening Assay Kit is designed for screening inhibitors of IL-17A:IL-17RA interaction. The IL-17RA[Biotin]-IL-17A Binding Assay Kit comes in a convenient 96-well format, with biotin-labeled IL-17RA, purified IL-17A, streptavidinlabeled HRP, and assay buffer for 100 binding reactions. The key to this kit is the high sensitivity of detection of biotin-labeled IL-17RA by streptavidin-HRP. Only a few simple steps on a microtiter plate are required for the assay. First, IL-17A is coated on a 96-well plate. Next, IL-17RA is incubated with IL-17A on the plate. Finally, the plate is treated with streptavidin-HRP followed by addition of an HRP substrate to produce chemiluminescence, which can then be measured using a chemiluminescence reader.

  • Background: IL-17A is a pro-inflammatory cytokine produced by TH17 cells that plays a key role in inflammation, autoimmunity, and host defense. It has been shown to stimulate the production of pro-inflammatory cytokines, namely IL-6 and IL-8, by synoviocytes. Due to its role as a mediator of inflammation, the IL-17 pathway has become a popular target for treating diseases that have a strong inflammatory component such as psoriasis, arthritis, Crohn's disease, and ankylosing spondylitis.
