ISRE Luciferase Reporter Lentivirus (JAK/STAT Signaling Pathway)

DESCRIPTION

The JAK/STAT pathway is activated by various cytokines and growth factors and plays a critical role in cell growth, hematopoiesis, and immune response. In mammals, there are four JAKs (JAK1, JAK2, JAK3 and TYK2) and seven STAT proteins. IFNα is a Type I interferon. Binding of IFNα to its receptor leads to the activation of JAK1 and TYK2, which in turn phosphorylate and activate STAT1 and STAT2. The phosphorylated STAT1 and 2 form a heterodimer and bind to IRF9/p48, forming a protein complex ISGF3. This complex translocates to the nucleus and binds to the ISRE (Interferon Stimulated Response Element) in the promoter region, thereby promoting transcription of interferon-inducible genes.The ISRE Luciferase Reporter Lentivirus are replication incompetent, HIV-based, VSV-G pseudotyped lentiviral particles that are ready to be transduced into almost all types of mammalian cells, including primary and non-dividing cells. The particles contain a firefly luciferase gene driven by multimerized ISRE response element located upstream of the minimal TATA promoter. After transduction, the activity of Type I interferon-induced JAK/STAT signaling pathway in the target cells can be monitored by measuring the luciferase activity.

  • Notes: Biosafety: The lentiviruses are produced with the third generation SIN (self-inactivation) lentivector which ensures self-inactivation of the lentiviral construct after transduction and integration into the genomic DNA of the target cells. None of the HIV genes (gag, pol, rev) will be expressed in the transduced cells, as they are expressed from packaging plasmids lacking the packing signal. Although the pseudotyped lentiviruses are replication-incompetent, they require the use of a Biosafety Level 2 facility. BPS recommends following all local federal, state, and institutional regulations and using all appropriate safety precautions

  • Formulation: The lentiviruses were produced from HEK293T cells in the medium containing 90% DMEM + 10% FBS.
