- Background: Isocitrate dehydrogenases are enzymes that catalyze the oxidative decarboxylation of isocitrate to 2-oxoglutarate. Mutations in isocitrate dehydrogenases 1 and 2 (IDH1 and IDH2) have been reported in a multitude of human cancers. While the wild type IDH1, produces α-ketoglutarate (α-KG) through the reduction of NADP+ to NADPH, the IDH1(R132H) mutant catalyzes conversion of α-ketoglutarate to 2-hydroxy-glutarate (2-HG) by oxidizing NADPH to NADP+. Genomic studies have identified the IDH1(R132H) mutation in various cancer types, including glioma, chondrosarcoma, and AML as well as a small percentage of prostate, lung and colon cancers.
