• Assay Conditions: Enzymatic activity was confirmed by an AmplexRed/Peroxidase coupling method using 1,5-diaminopentane as a substrate. Purified LOXL4 (0.25-1 ng/µl) was incubated with 3 mM 1,5 diaminopentane, 0.4 mM AmplexRed and 0.2 unit/µl peroxidase in 100 µl of the assay buffer (1.2 M Urea, 50 mM borate pH 8.8) for 15 min at 30°C. Fluorescence intensity is measured every 7 min at 530ex/590em.
