Since 1988 we have a facility in the Netherlands Cancer Institute to synthesize peptides for researchers in our institute outside.
We use Fmoc-solid phase chemistry and in our facility we have three robots (a Prelude syntheziser and two Syro synthesizers). One of the Syro’s can synthesize 48 peptides simultaneously. The other robot can synthesize in one run 3 times 96 peptides in small amounts (2 to 4 µmol) for screening purposes. The Prelude is used for larger amounts of peptides.
Peptides are routinely synthesized in the range of 5-100 amino acids; in an amount of 2-200 µmol (larger amounts are possible). All peptides are analyzed by LCMS and if needed purified by
RP-HPLC. For analytical purposes the peptide facility has a Waters liquid chromatography electro spray mass spectrometer at its disposal. More than 5,000 peptides are synthesized annually.
Besides “straight forward” peptides we can also synthesize special peptides with unnatural amino acids, ubiquitin and its derivatives, biotinylated-, acetylated-, phosphorylated peptides, peptides with a side chain, custom dye labeled and UV cleavable peptides.
