The Whitehead Proteomics Core Facility provides proteomics resources to Whitehead and MIT community as well as other institutions. We utilize high resolution mass spectrometry and liquid chromatography for protein identification, comparative proteomics and quantitative proteomics as well as the identification of post-translational modifications. Our technology uses “bottom-up” proteomics technology where proteins are digested with trypsin and the resulting peptides are chromatographicly separated and analyzed by tandem mass spectrometry on an Orbitrap Elite mass spectrometer. Protein identification and quantitative information are obtained through using database search engines and species specific databases. The results are presented in a spreadsheet format using commercially available viewers.
The facility performs both quantitative proteomics analysis, post translational modification identification and simple protein identifications. We work with both gel-based and solution-based samples, perform two dimensional chromatographic separations, and stable isotope labeling methods including SILAC, TMT and iTRAQ.
